Vabadin® 10 mg belongs to the pharmacological group of anti-sclerotic drugs and is a hypolipidemic drug that affects the synthesis of lipoproteins (complex proteins that carry cholesterol) and regulates the level of cholesterol, triglycerides (fatty esters) and phospholipids (fat-like substances) in the blood plasma.
Indications Vabadin 10 mg
This drug is used in the complex therapy of cardiovascular diseases that are caused by or accompanied by a disorder of lipid (fat) metabolism in the body: increased cholesterol levels in the blood (hypercholesterolemia) of all types, as well as pathologically high levels of lipids or lipoproteins (dyslipidemia).
In addition, Vabadin® 10 mg (synonyms - Simvastatin, Vazilip, Zocor, Simvacard, Simvor, Simgal) is prescribed to prevent the development of cardiovascular pathologies associated with atherosclerosis.
Release form
Release form Vabadin® 10 mg - film-coated tablets in a blister pack of (14 pieces), in a cardboard pack - 2 blister packs. One tablet of Vabadin® 10 mg contains 10 mg of simvastatin, as well as excipients, including lactose monohydrate.
Pharmacodynamics
The active substance of the drug Vabadin® 10 mg - the statin simvastatin (simvastatin) - is metabolized in the body with the formation of free oxycarboxylic acids, which suppress the production of the enzyme (3-hydroxy-3-methyl-glutaryl-coenzyme A reductase), necessary for the liver to produce cholesterol, and thereby reduces the concentration of total cholesterol in the blood. The concentration of cholesterol in the blood largely depends on the rate of its capture by special cells (atherogenic low-density lipoproteins or LDL and very low density - VLDL), which transfer cholesterol and triglycerides to the cells of all tissues, including the walls of blood vessels.
Vabadin® 10 mg also stimulates the formation of cellular receptors for "bad" cholesterol by high-density lipoproteins (HDL), which can remove cholesterol from cells and transfer it to the liver for decomposition by oxidation into bile acids. As a result of these biochemical processes, cholesterol deposition on the walls of blood vessels decreases, i.e. the risk of atherosclerosis, and in ischemic heart disease (IHD) - the threat of myocardial infarction.
Pharmacokinetics
After oral administration, the drug is well absorbed in the gastrointestinal tract, up to 95% of the active substance binds to plasma proteins. The pharmacologically active metabolite is formed in the liver, and its maximum concentration in the blood plasma is achieved 1-2 hours after taking the drug.
Most of the drug Vabadin® 10 mg is excreted unchanged through the intestines, the rest - with urine. The average time of complete elimination of the drug from the body is 96 hours.
Dosing and administration
The dosage and duration of treatment are determined by the doctor. The maximum daily dose of Vabadin is 80 mg, taken once a day - in the evening. The tablets are taken orally whole, with 200 ml of water.
In case of high risk of cardiovascular pathology – for prevention purposes – 20-40 mg of the drug is prescribed. If necessary, the dose can be gradually increased (the interval between dose increases is at least four weeks).
If concomitant treatment with bile acid secretion enhancing drugs is administered, Vabadin should be taken 4 hours after or 2 hours before taking these drugs.
Use of the maximum daily dose of Vabadin® 10 mg requires regular monitoring of blood lipid levels, liver function, and the general well-being of the patient.

Use Vabadin 10 mg during pregnancy
The use of Vabadin® 10 mg during pregnancy and breastfeeding is contraindicated. Women of childbearing age are advised to use reliable contraception while taking the drug. The drug is not prescribed to children and adolescents under 18 years of age.
Contraindications
Contraindications to the use of Vabadin® 10 mg include: individual hypersensitivity to the components of the drug; exacerbation of liver disease; lactase deficiency (impaired absorption of milk protein).
Side effects Vabadin 10 mg
The use of this medicine may cause the following side effects: headache and dizziness; fever; nausea, vomiting; skin rash, itching, urticaria, hair loss; shortness of breath, flushing of the face and upper body; pain in the upper abdomen, muscles and joints; bowel disorder, flatulence; paresthesia and muscle spasms; pancreatitis, hepatitis, jaundice.
Overdose
Overdose of the drug leads to more pronounced side effects.
Methods for eliminating the consequences of a drug overdose include gastric lavage, taking enterosorbents, and, if necessary, symptomatic treatment.

Interactions with other drugs
It is contraindicated to combine the use of Vabadin® 10 mg with drugs that inhibit the CYP3A4 enzyme – erythromycin, nefazodone, itraconazole and ketoconazole.
Vabadin® 10 mg should not be prescribed simultaneously with a drug of the same pharmacological group - the lipid-lowering agent gemfibrozil.
Concomitant administration of drugs such as cyclosporine, danazol and niacin requires mandatory adjustment of the dosage of Vabadin® 10 mg to a maximum daily dose not exceeding 10 mg. Special note: during treatment with Vabadin® 10 mg, consumption of grapefruit juice is contraindicated, as this juice leads to an uncontrolled increase in the effect of the drug.
Storage conditions
The drug should be stored in a dry place, away from direct sunlight, at a temperature of up to +25°C.

Special instructions
Elderly patients, as well as patients with kidney pathologies, thyroid hormone deficiency (hypothyroidism), alcohol dependence and diseases of the muscular system, Vabadin® 10 mg is prescribed with caution. This also applies to those whose work is related to driving vehicles and mechanisms.
Shelf life
The shelf life of the drug is 3 years.
