Pharmacodynamics

The drug Nise-gel has a local anesthetic and anti-inflammatory effect. The active component of the drug - nimesulide - is a selective competitive reversible inhibitor of cyclooxygenase 2. The substance is able to reduce the content of some prostaglandins, which are present in the foci of inflammatory processes, as well as in the pathways that conduct pain impulses in the spinal cord. All this leads to the appearance of analgesic and anti-inflammatory effects.

Pharmacokinetics

Local application of Nise-gel causes low absorption of active substances, which leads to very low concentrations of the components of the drug in the circulatory system. The maximum amount of the active substance after a single use of the gel is observed approximately twenty-four hours later. The concentration of nimesulide in this form of its application is three hundred times lower than when using its oral forms. The main metabolite of the active substance, 4-hydroxynimesulide, is not detected in the blood.
