Lavex is used for erectile dysfunction (impotence), i.e. when a man's penis, during sexual arousal, does not reach sufficient volume and hardness for full sexual intercourse.
Indications Lavex
Lavex is prescribed for erectile dysfunction in men. This drug is prescribed when lower doses of sildenafil have not produced the desired result.
The main active ingredient of the drug is sildenafil, which has become very popular (better known as Viagra). Restoration of erectile function occurs due to improved blood circulation in the penis, which leads to a natural erection during sexual arousal. This occurs due to the release of nitric oxide in the cavernous bodies of the penis during sexual arousal, then guanylate cyclase is activated and the concentration of cGMP increases, causing relaxation of the smooth muscles in the cavernous bodies, which promotes blood flow. The drug begins to act only under the condition of sexual stimulation. The maximum concentration of the drug in the blood is observed on average an hour after administration. Lavex is taken on an empty stomach, in this case the absorption rate will be higher. Elderly people need to take the drug, starting with a dose of 25 mg, then, taking into account the condition and effectiveness, the dose can be increased to 50 or 100 mg. It is also necessary to start taking Lavex with 25 mg for those who have severe liver and kidney diseases, then the dose can be increased.

Pharmacodynamics
Lavex is an oral medication that is designed to restore erection in men. The drug effectively restores erectile function and provides a natural response to sexual arousal. The process of physiological erection involves the release of nitric oxide in the corpus cavernosum during sexual arousal. Nitric oxide activates the enzyme guanylacitase, which increases the level of cGMP, increases blood circulation in the penis, and relaxes smooth muscles. Sildenafil is a strong selective inhibitor (antidepressant) that enhances the relaxing effect in the tissues of the corpus cavernosum. Sildenafil, the main substance of Lavex, exhibits its pharmacological effect only with appropriate sexual stimulation.
Pharmacokinetics
Lavex is quickly absorbed after oral administration. The maximum concentration of the drug in the blood is achieved on average about an hour after taking the tablet, provided that it was drunk on an empty stomach, otherwise the absorption of the drug is impaired. The half-life of saldenafil is about four hours. Sildenafil is excreted from the body in the form of metabolites mainly with feces (about 80% of the dose taken) and urine.
Studies on healthy volunteers aged 65 and over showed that the excretion of sildenafil was reduced and the concentration of the active substance was 90% higher than in younger volunteers aged 18 to 45 who took part in the experiment. Among volunteers with mild or moderate renal insufficiency, the pharmacokinetics did not change, and with severe renal insufficiency, the excretion of Lavex from the body was reduced.
Dosing and administration
Lavex is taken when drugs containing lower doses of sildenafil have not brought the expected result. The drug is prescribed orally, one tablet per day (70 mg) one hour before sexual intercourse.
In case of liver or kidney dysfunction, Lavex is prescribed in smaller doses. It is necessary to adhere to the appropriate dosage.

Use Lavex during pregnancy
Lavex is not recommended for use during pregnancy and breastfeeding, since clinical trials in this area have not been conducted.
Contraindications
Lavex should not be used by men under 18 years of age. Also, the drug is not prescribed if there is hypersensitivity to the components of the drug.
The drug is contraindicated for people who should not have an active sex life (for cardiovascular diseases).
Use the drug with caution in case of anatomical changes of the penis. Taking Lavex may be accompanied by dizziness, visual impairment, so it is necessary to refrain from driving a car until the individual reaction of the body to the drug is established. You cannot take Lavex simultaneously with nitrates or other NO.

Side effects Lavex
Lavex should be taken approximately one hour before sexual intercourse, the recommended dose is 50 mg. Taking into account the individual reaction of the body to the drug, the dose can be increased to 100 mg or reduced to 25 mg. Lavex is taken only once a day, as indicated above, simultaneous food intake affects the effectiveness of the drug, the desired effect will be achieved much later (approximately 2-3 hours after taking the tablet).
Lavex can provoke angina, migraine, tachycardia, heart failure, cardiac arrest, cerebral vascular thrombosis, cardiomyopathy, atrioventricular block, arterial hypotension.
From the digestive system, stomatitis, dyspepsia, vomiting, xerostomia, and rectal bleeding may occur.
The central nervous system may react with headache, ataxia, tremor, dizziness, insomnia, depression, hyporeflexia, hypoesthesia.
From the respiratory system, laryngitis, urticaria, cough, sinusitis, bronchitis, asthma, and pharyngitis may occur.
Various allergic reactions are also possible – herpes simplex, dermatitis, ulcers, itching.
Taking Lavex may cause conjunctivitis, eye pain, visual impairment, cataracts, bleeding, cystitis, photophobia, urinary incontinence, swelling of the genitals, anorgasmia, facial hyperemia, and swelling of the nasal mucosa.

Overdose
Clinical trials on healthy volunteers, exceeding the recommended doses of the drug resulted in a feeling of heat, dizziness, headache, hypotension, and visual impairment. In case of overdose, standard treatment is carried out in such cases. Treatment with dialysis (a procedure that partially takes over the functions of the kidneys) does not increase the elimination of sildenafil from the body, since it actively binds to plasma proteins and is not excreted in the urine.
Interactions with other drugs
It is not recommended to use Lavex in combination with other drugs intended for the treatment of erectile dysfunction; the safety and effectiveness of this type of interaction has not been studied.
You cannot take saldenafil and ritonavir at the same time. Studies have shown that the elimination of sildenafil is reduced when taken simultaneously with CYP3A4 inhibitors - erythromycin, ketoconazole, cimetidine.
Administration of an antacid (magnesium/aluminum hydroxide) had no effect on the bioavailability of sildenafil.
In patients with arterial hypertension taking amlodipine, no signs of interaction with sildenafil were found.
CYP 2C 9 inhibitors (warfarin, tolbutamide), CYP 2D6 (antidepressants), thiazides, thiazide-like diuretics, ACE inhibitors, calcium antagonists, beta-adrenergic receptor blockers, CYP 450 metabolism inducers (barbiturates, rifampicin) did not affect the pharmacokinetic properties of the drug.

Storage conditions
Lavex should be stored out of reach of children. The drug should be protected from direct sunlight and stored at a temperature not exceeding 25 0 C.
Special instructions
Before taking Lavex, you must undergo a cardiological examination.
After the drug went on sale, cases of serious heart problems were recorded after taking the drug - angina pectoris, myocardial infarction, gastric arrhythmia, hemorrhagic stroke, sudden coronary death. These side effects developed in people with heart disease. All of these cases occurred mainly during or immediately after sexual intercourse. The drug is not prescribed to patients who have recently suffered a stroke or myocardial infarction, since the effect of the drug in such conditions has not been studied.
Also, the drug should be taken with caution by people with an exacerbation of a stomach or duodenal ulcer.
At the beginning of taking Lavex, it is necessary to establish the individual reaction of the body to the drug; you should refrain from driving a car or other complex mechanisms.
Shelf life
Lavex retains its properties for two years from the date of manufacture, subject to storage conditions. The drug cannot be used after the expiration date.
