The drug belongs to the category of non-steroidal anti-inflammatory drugs.
Inhibition of the cyclooxygenase enzyme stops the formation of prostaglandins. The drug does not inhibit COX-1, and the increase in COX-2 activity is a response to the influence of anti-inflammatory factors synthesizing the accumulation of prostaglandins, especially E2. When using therapeutic doses, the synthesis of constitutional prostaglandins is not disrupted and there is no negative effect on physiological processes in tissues (stomach, intestines, duodenum).
Indications Celecoxib-Norton
The drug is prescribed for adult patients with connective tissue diseases.
Indications for use of Celecoxib:
- Osteoarthritis is a pathological process of wear and tear of the articular apparatus of the extremities. It occurs as a result of injuries, damage or hereditary predisposition. As a rule, it affects the knee joints of women, progresses quickly, causes severe pain, has several types and stages of development.
- Rheumatoid arthritis is a systemic inflammatory disease affecting connective tissues. It is characterized by progressive erosive and destructive processes. The disease is autoimmune, as it occurs when the immune system is disrupted. It occurs in 2% of the world's population and affects symmetrical joints of the feet and bones.
Release form
The antirheumatic drug is available in capsule form for oral use. The chemical name of the drug is diaryl-substituted pyrazole. The release form allows you to calculate the number of tablets needed to complete a full course of treatment.
Each capsule contains 100 mg or 200 mg of the active substance - celecoxib. Gelatin capsules, hard, with a green 100 mg or pink 200 mg cap containing white crystalline powder. Excipients: colloidal silicon anhydrous, calcium phosphate dibasic, corn starch, lactose, sodium starch glycolate and other substances.
Pharmacodynamics
The medicinal efficacy of Celecoxib is based on its mechanism of action. Pharmacodynamics indicate that Celecoxib belongs to the category of non-steroidal, antirheumatic and anti-inflammatory drugs. The analgesic effect is based on antipyretic activity.
The drug suppresses the synthesis of prostaglandins by selectively inhibiting the enzyme cyclooxygenase, which is involved in inflammatory processes. Tablets are recommended to be taken only with appropriate indications and medical permission.
Pharmacokinetics
After oral administration, Celecoxib Norton is rapidly absorbed in the digestive tract, with maximum plasma concentration occurring after 3 hours. If the tablets are taken with food, this slows down absorption by 1-2 hours. Pharmacokinetics are proportional and linear to the dose taken. Plasma protein binding is 95-97%.
The active substance is metabolized in the liver by hydroxylation, oxidation and glucuronidation. It is excreted mainly in the bile as metabolites. About 3% is excreted unchanged in the urine and feces. With repeated use, the half-life is 8-12 hours, and the concentration in the blood plasma is achieved within 5 days.
Dosing and administration
The medication is intended for oral use. The method of administration and dosage of Celecoxib depend on the indications for its use. Doctors recommend the minimum effective dose with a shorter course of treatment.
- Osteoarthritis – 100 mg twice daily or 200 mg once.
- Rheumatoid arthritis – up to 200 mg twice a day (dosage depends on the severity of the patient’s condition).
If the drug is prescribed to patients over 65 years of age, the daily dose should not exceed 200 mg. The duration of therapy depends on the clinical condition of the patient and, as a rule, does not exceed 15 days. To prevent irritation of the esophageal mucosa, capsules are recommended to be swallowed without chewing, with plenty of liquid.
Use Celecoxib-Norton during pregnancy
The use of the drug in the first trimester of pregnancy is contraindicated. This is due to the increased risk of developing deformities and pathologies (heart defects, cleft palate). The use of Celecoxib during pregnancy is possible only with appropriate medical indications, when the benefit to the mother is higher than the potential risks to the fetus. Contraindications are related to the fact that the active component penetrates the placental and BBB barrier.
If the capsules are taken in the third trimester, this may provoke postmaturity and a significant weakening of labor. The use of the drug during lactation in moderate dosage is possible. When taking increased doses, breastfeeding should be stopped.
Contraindications
The use of Celecoxib is possible only with appropriate indications and medical permission. This is due to the fact that the drug causes negative reactions from many organs and systems.
Contraindications for use:
- Individual intolerance to the active substance and auxiliary components.
- Predisposition to the development of allergic reactions (urticaria, rhinitis, bronchospasms) when using non-steroidal anti-inflammatory drugs and aspirin.
- Pregnancy and lactation period.
- Patients under 18 years of age.
Side effects Celecoxib-Norton
If a medicine is taken without following the doctor's recommendations and self-selecting the dosage, it causes a number of side effects.
Celecoxib may cause problems such as:
- Flatulence, diarrhea, constipation, abdominal pain.
- Various disorders of the nervous system (headaches, insomnia, dizziness).
- Upper respiratory tract infections.
- Back pain.
- Peripheral edema.
- Rhinitis and pharyngitis.
Side effects of Celecoxib are reduced when using the minimum effective dose during a short course of treatment. If the tablets are prescribed to elderly patients, the side effects are increased and pronounced with prolonged use. With prolonged therapy, regular monitoring of the patient's condition is provided.
Overdose
Failure to comply with the rules for using the medicine provokes various adverse symptoms. Overdose occurs when the dose prescribed by the doctor is exceeded.
Signs of overdose:
- Intoxication of the body, which is accompanied by vomiting, painful sensations in the epigastric region, nausea, tinnitus, decreased visual and hearing acuity, and dizziness.
- Severe overdoses may cause impaired thinking, drowsiness, tremors of the extremities, shortness of breath, clouding of consciousness, dehydration, and hyperthermia.
To eliminate the above symptoms, gastric lavage is performed. Depending on the acid-base and electrolyte balance, infusion of sodium lactate, sodium bicarbonate and sodium citrate solutions is prescribed.
Interactions with other drugs
The use of any medicine with other medications is possible only with the appropriate medical recommendations. Interaction with other drugs - anticoagulants, including aspirin, is contraindicated, as this can enhance their effect.
Simultaneous use with Theophylline, Digoxin, Warfarin, Glibenclamide and antacids is possible. Such interaction does not cause clinically significant disorders. But do not forget that non-steroidal anti-inflammatory drugs reduce lithium clearance, which increases its level in blood plasma and toxicity.
Storage conditions
The medicinal effect of the drug largely depends on storage conditions. Celecoxib-Norton must be kept in the original packaging, protected from sunlight, moisture and out of the reach of children.
The storage temperature should not exceed 25°C. If these recommendations are not followed, the medicine loses its properties and may cause uncontrolled side effects in many organs and systems.
Shelf life
The mechanism of action of the drug depends on its validity period. This is due to the fact that the use of expired drugs is unsafe for the patient's body and causes many pathological symptoms. Celecoxib is taken for 36 months from the production date, which the manufacturer indicates on the packaging of the tablets. After this period, the capsules must be disposed of.
