Pharmacodynamics

Tamsulosin is a substance from the category of drugs specifically blocking the activity of postsynaptic α1-adrenergic receptors located inside the smooth muscles of the prostate, the neck of the ureter and the prostatic section of the urethra. After blocking the activity of α1-adrenergic receptors under the influence of tamsulosin, there is a weakening of the tone of the muscles of the prostate, urethra and the prostatic area of the urethra, resulting in an improvement in urinary processes. Along with this, there is a decrease in the symptoms of filling and emptying of urea with prostate adenoma associated with detrusor hyperactivity and increased muscle tone of these organs.

The effect of tamsulosin on adrenoreceptors of the α1A subtype is 20 times stronger than that of the adrenoreceptors of the α1B subtype within the smooth muscle of the vessels. Tamsulosin, which remains unchanged in its unaltered state, is excreted through the kidneys together with its metabolic products. The half-life of a 1-fold oral dose (0.4 mg) of the drug after a meal is 10 hours. After reusable methods of drugs, these values reach 13 hours.

Dosing and administration

Capsules of the drug are consumed orally, immediately after the end of breakfast. The medicine should be washed down with plain water (0.1-0.2 l). During the day, take 400 μg of the substance (contained in the 1st capsule).

Contraindications

The main contraindications:

  • presence of strong sensitivity to tamsulosin or other components of the drug;
  • insufficiency of the liver, which has a severe form;

Overdose

The values of blood pressure and heart rate can be normalized after placing the patient in a horizontal position. If this method does not work, prescribe the use of drugs that increase the blood volume inside the vascular bed. If necessary, vasoconstrictor medicines may be used. In addition, during therapy, kidney function should be monitored. Sorbents can also be used and gastrointestinal cleaning (gastric lavage and consumption of laxatives).

Since tamsulosin binds heavily to the protein, hemodialysis procedures will be ineffective.

Interactions with other drugs

Combination of the drug with nifedipine, atenolol or enalapril does not lead to the development of any therapeutic interaction.

The combination with cimetidine causes a slight increase in plasma indices of tamsulosin, and in the case of admission with furosemide, a decrease in this value is observed. With such a medicamentous combination, the dosage of Omnik is not required to change, because the plasma level of its acting element remains within the limits of the norm.

Combination of a drug with diclofenac or warfarin can increase the rate of excretion of its active element.

Simultaneous use of α1-adrenergic receptors together with other antagonists can lead to a decrease in blood pressure.
