Pharmacodynamics

The active element of the drug is tamsulosin, which is included in the category of substances specifically and competitively blocking the activity of postsynaptic α1-adrenergic receptors having a particular affinity for their α1A, as well as α1D subtypes (they are responsible for the relaxation of the smooth muscle tissue of the prostate, the prostatic area of the urethra, and neck of the urethra).

An internal portion of tamsulosin, 400 μg, increases the rate of the urinary process and weakens the tone of the urethra and prostate muscles, resulting in an improvement in urinary outflow and a decrease in the negative symptoms associated with the unstable function of the urea. This effect reduces the signs of obstruction and irritation caused by prostate adenoma, which persists even with prolonged therapy.

Like the other α1A-adrenergic receptors, tamsulosin can reduce the values of AD, weakening the peripheral resistance, but it should be noted that the daily use of 400 μg of the substance did not lead to a clinically significant decrease in blood pressure.

Since the tablets of the preparation have a slow release type of the active ingredient, tamsulosin is secreted slowly, demonstrating exposure with minimal variation in plasma medication values over the next 24 hours.

Pharmacokinetics

Absorption of tamsulosin takes place inside the intestine, where about 57% of the consumed portion of the drug is absorbed. The intake of food does not affect the degree of absorption.

Contraindications

The use of medication can trigger the emergence of a variety of adverse events:

• vomiting, diarrhea, constipation, and nausea;
• headaches, asthenia, dizziness, fainting (occasionally);

• priapism (single) or an ejaculation disorder.

Overdose

Interactions with other drugs

Combination with warfarin, simvastatin, and in addition diazepam, amitriptyline, diclofenac and propranolol, as well as trichloromethiazide, glibenclamide and chloromadinone does not lead to changes in the free drug fraction within the plasma (in vitro), just as tamsulosin does not affect the values free fraction of propranolol, chloromadinone and diazepam with trichloromethiazide.

The combination with other drugs from the category blocking the activity of α1-adrenergic receptors can cause a potentiation of the antihypertensive effect.

Combination of drugs with cimetidine can slightly increase the value of tamsulosin inside the plasma, while the combination with furosemide causes their decrease. But with such changes, Omnic Okasa portions need not be adjusted, because the plasma level of its active substance remains within acceptable limits.

Performed in vitro tests of metabolic transformations carried out inside the liver, did not record the development of the interaction between tamsulosin and glibenclamide, finasteride, and besides, amitriptyline and salbutamol.

Combination with diclofenac or warfarin slightly increases the rate of excretion of tamsulosin.

Simultaneous use of the drug with atenolol, enalapril, theophylline or nifedipine does not lead to the development of any therapeutic interactions.
