Lerivon is a tetracyclic drug that belongs to the piperazine-azepine group of antidepressants.

The drug helps block α2-adrenoreceptors, and in addition increases the indicators of the norepinephrine mediator inside the synapse gap. Potentiated noradrenergic transmission of neuronal impulses helps increase brain activity. This is how the anxiolytic effect of the therapeutic agent is mediated.

Activation of α-adrenergic and H1-histamine endings leads to the development of an intense sedative effect of the drug.

It is used for depressions of various origins.

Release form

The medicinal substance is released in tablets, 20 pieces per cell plate.

Dosing and administration

The medication should be taken orally, without chewing the tablet, and washed down with plain water.

At the beginning of the course, an adult needs to take 30 mg of the substance. Taking into account the clinical effect, the portion is adjusted. The size of the maintenance dosage should be lower than the initial one. A maximum of 60-90 mg of the drug can be administered per day.

Elderly people are initially prescribed a similar dose of the drug. In order for the medicinal effect to occur, smaller doses are required; their titration is performed by the attending physician during the course of the treatment cycle.

It is recommended to use Lerivon in the evening, before bed, because it has a strong sedative effect.

On average, the therapeutic course lasts 4-6 months. Antidepressant treatment is continued for several more months after the clinical picture has improved. It is forbidden to suddenly stop taking the drug, because this increases the likelihood of withdrawal syndrome.

Use Lerivona during pregnancy

There is only limited information regarding the safety of using the drug during lactation or pregnancy. Before prescribing Lerivon, the doctor should evaluate all possible risks and benefits from taking it.

Contraindications

Among the contraindications:

  • manic syndrome;
  • appointment for therapy for depression in people with severe liver dysfunction, because the drug can accumulate, which can lead to intoxication;
  • appointment to people with diagnosed intolerance to elements of the drug.

Side effects Lerivona

Often, side effects include disorders associated with the functioning of the central nervous system: convulsive readiness accompanied by hyperkinetic syndrome, and in addition to this, hypomania and CNS.

At the initial stage of drug administration, a tendency to develop various arrhythmias or bradycardia may be observed.

Due to the pharmacokinetics of the drug, jaundice may sometimes develop or the activity of intrahepatic enzymes may increase.

Granulocytopenia or agranulocytosis may also be observed.

In some people, the appearance of allergic rashes or the development of arthralgia was observed.

Overdose

In case of poisoning with the drug, prolonged sedation is observed. Hypotension, cardiac arrhythmia, convulsive readiness and external respiration disorders appear less frequently.

The drug has no antidote. In case of overdose, symptomatic, supportive and detoxifying actions are performed (this includes gastric lavage in the first few hours after taking the drug). If severe disorders of vital activity are observed, this parameter is constantly monitored.

Interactions with other drugs

The drug enhances the effect of MAOIs, so they are not used for at least 14 days after the end of treatment with tricyclics.

The drug contributes to the disruption of metabolic processes of indirect coagulants, which is why when combining them it is necessary to regularly monitor the function of the coagulation system.

Lerivon does not affect the pharmacokinetic characteristics of antihypertensive agents (clonidine and hydralazine with guanethidine, as well as betanidine and methyldopa with propranolol).

Alcoholic beverages potentiate the drug's inhibitory effect on the central nervous system, which is why patients should refrain from drinking them during therapy.

Storage conditions

Lerivon must be stored at temperatures in the range of 2-30°C.

Shelf life

Lerivon can be used within a 5-year period from the date of sale of the therapeutic substance.

Application for children

The drug cannot be prescribed in pediatrics.
