Denebol gel is an antirheumatic NSAID. It belongs to the coxibs group.
Indications Denebol gel
The medicine is indicated for the treatment of osteoarthritis (in acute or chronic form), periarthritis, rheumatoid arthritis, and also bursitis with thrombophlebitis and inflammatory process in the area of tendons. Along with this, it is also used for osteoarthrosis and osteochondrosis, damage to the musculoskeletal system, lumbago and neuralgia. It is also used for inflammation in the area of muscles, ligaments and tendons with joints, developing after injuries (for example, as a result of overexertion, stretching or impact).
Release form
It is produced in gel form in 30 g tubes. One pack contains 1 tube of gel.
Pharmacodynamics
The active component of the gel is rofecoxib (NSAID) - a pronounced selective inhibitor of COX-2. When used locally, it has an antipyretic, anti-inflammatory and analgesic effect. Activation of COX-2 occurs as a reaction to inflammation - as a result, accumulation and synthesis of inflammatory conductors begins (among them Pg E2), which causes swelling, inflammation, and pain. The anti-inflammatory properties of rofecoxib are due to the suppression of the process of PG synthesis by slowing down the activity of COX-2.
Medicinal concentrations of the drug do not inhibit COX-1, which allows the drug not to affect PGs that are bound as a result of the active action of COX-1. That is why it does not interfere with the natural physiological processes occurring inside the tissues (this especially concerns the gastrointestinal tract, as well as platelets).
Flaxseed oil is a demulsified agent with a softening effect, which is a component of locally applied drugs. It contains α-linoleic acid (it is very important for the functioning of the human body).
Menthol has cooling properties and has a therapeutic effect when applied locally to the skin. This component stimulates "cold" endings and normalizes the movement of Ca ions along the membranes of neurons.
Methyl salicylate is a rubefacient for local use in arthralgia, myalgia and soft tissue injuries. In addition, it has anti-inflammatory and analgesic properties, as it is able to normalize the COX enzyme.
Pharmacokinetics
The absorption of the gel applied to the skin into the systemic bloodstream is quite slow. Even the use of the drug in large quantities does not increase the plasma concentration of the active component.
Dosing and administration
The skin surface should be treated with the gel 3-4 times a day (light rubbing). The dosage depends on the area of the injured area. For example, about 2-4 g of gel will be enough to treat an area of approximately 400-800 cm. For children, 1-2 g of the medicine will be enough.
After the procedure, you need to wash your hands. The total duration of the treatment course depends on the effectiveness of the drug, as well as the indications.
Use Denebol gel during pregnancy
There is no data on the use of the drug by pregnant women, but in any case, rofecoxib is only permitted to be used in cases of high probability of benefit to the mother and low risk of negative impact on the fetus.
There is no information on the penetration of the active ingredient into breast milk, so it is necessary to temporarily stop breastfeeding during the period of using the drug.
Contraindications
Contraindication is intolerance to rofecoxib or other components of the drug.
Interactions with other drugs
With local use of the recommended dosage, the systematic absorption of the active components will be lower than the probability of negative drug interactions. However, with excessive local use of methyl salicylate, it is possible for it to be absorbed through the skin and an interaction similar in manifestation to warfarin may develop.
Shelf life
Denebol gel is suitable for use for 2 years from the date of release of the drug.
