Aleric is an antihistamine drug containing the element loratadine. This component is a derivative of piperidine; it is a 2nd generation antiallergic drug that exhibits long-term antihistamine activity. Aleric selectively blocks the activity of peripheral H1-histamine receptors.

The drug has antipruritic, antiallergic and antiexudative effects; at the same time, loratadine demonstrates an anti-edematous effect and strengthens capillary strength. [ 1 ]

Indications Alerica

It is used for rhinitis of allergic etiology (this includes its seasonal and year-round forms), and also for conjunctivitis, which also has an allergic origin.

It can be prescribed to people with dermatitis of an allergic nature and idiopathic urticaria (in the chronic phase).

Release form

The medicine is produced in tablets - 7 pieces (1 cell package inside a pack) or 10 pieces (3 cell packages inside a box).

Pharmacodynamics

Loratadine is almost not synthesized with histamine endings of the central nervous system, and also has no affinity for other endings of biogenic amines. The size of the molecule in combination with a high rate of protein synthesis almost completely prevents the penetration of loratadine through the BBB. [ 3 ]

The medicinal effect of the drug is noted after 1-3 hours, reaching the highest effective values after 8-12 hours from the moment of administration. The effect of the drug lasts for 24-48 hours. [ 4 ]

Pharmacokinetics

When administered orally, loratadine is well absorbed in the gastrointestinal tract. The Cmax level in plasma is recorded after 1-2 hours from the moment of administration.

Approximately 99% is synthesized with intraplasmic protein. Equilibrium indices in plasma are observed by the 5th day of drug administration.

The processes of loratadine metabolism are realized inside the liver with the help of the structure of hemoprotein P450 (this includes CYP 34A with CYP 2D6). During metabolism, a metabolite with therapeutic activity is formed, as well as some inactive elements.

The half-life is in the range of 7.5-11 hours; the active element together with metabolic fragments is excreted mainly in the urine; a small part is excreted in the feces.

Loratadine does not accumulate in the body even with prolonged use. The drug is excreted in breast milk.

Dosing and administration

Aleric should be taken orally, without reference to food intake. If necessary, the tablets can be divided. The daily dose is taken in 1 dose (the drug should be used at the same time of day).

The duration of the therapeutic course and the size of the drug doses are selected by the attending physician, who takes into account the personal characteristics of the patient and the intensity of the manifestations of the pathology.

A child weighing over 30 kg and an adult are given the medicine in a daily dose of 10 mg (equivalent to 1 tablet of the medicine).

For a child under 12 years of age weighing less than 30 kg, the medication is prescribed in a daily dosage of 5 mg (equivalent to 0.5 tablets).

Treatment usually lasts for 10 days. If necessary, therapy can be extended, taking into account the effectiveness of the drug and its tolerability by the patient.

If the patient requires an epidermal allergy test, the use of Aleric must be stopped 7 days before this procedure.

Use Alerica during pregnancy

Loratadine should not be used during pregnancy.

If lactating women need to take the medication, they should stop breastfeeding for the duration of treatment. Breastfeeding can be continued for at least 3-5 days after stopping the medication (complete excretion of loratadine from the body occurs within 60-120 hours).

Contraindications

Main contraindications:

  • strong personal sensitivity to loratadine and other components that make up the drug;
  • glucose-galactose malabsorption;
  • galactosemia;
  • lactose intolerance.

It is prescribed with great caution to people with severe renal dysfunction (this includes cases of acute renal failure and situations with CC values below 30 ml per minute - in this case, the dose of the medication must be adjusted) and to the elderly.

Side effects Alerica

The medication is often tolerated without complications.

Occasionally, mainly in people with severe intolerance, nausea, increased fatigue, xerostomia, drowsiness and headaches may occur.

In addition, heart rhythm disturbances, epidermal rashes, alopecia, syncope, liver dysfunction and anaphylaxis may occur.

Overdose

The use of large doses of loratadine causes headaches, tachycardia and drowsiness, as well as loss of consciousness.

Conscious patients (if less than 2 hours have passed since the drug was taken) should undergo gastric lavage and take enterosorbents. In case of loratadine poisoning, the patient should be under medical supervision. Symptomatic measures are taken if necessary.

In case of intoxication with Aleric, hemodialysis will be ineffective.

Interactions with other drugs

It is prohibited to use loratadine in combination with substances containing ethanol, as well as with alcoholic beverages.

Administration of the drug together with macrolides, antifungals (imidazole derivatives with triazoles) and cimetidine may cause an increase in plasma levels of loratadine.
