Tamsulostad

The urological drug Tamsulostad is used for benign prostatic hyperplasia. The drug is an alpha 1- adrenergic receptor antagonist.
Indications Tamsulostada
Tamsulostad can be used to correct functional disorders of the urinary system caused by benign prostatic hyperplasia.
Release form
Tamsulostad is available as modified-release capsules.
The capsules are packed in blister strips, 10 pieces in each strip. The cardboard pack contains three strips, which is equivalent to 30 capsules of the drug.
The active ingredient is tamsulosin hydrochloride, the content of which in one capsule is 0.4 mg.
Pharmacodynamics
Tamsulostad is an alpha 1 -adrenoreceptor antagonist. The drug voluntarily inhibits postsynaptic alpha 1 -adrenoreceptors located in the smooth muscle structures of the prostate gland, bladder neck and prostatic portion of the urethra. This action causes a decreased tone of smooth muscle fibers, which affects the facilitation of urination. At the same time, signs of compression and irritation caused by benign prostatic hyperplasia disappear.
It has been observed that the therapeutic effect begins to manifest itself approximately 14 days after the first dose of Tamsulostad.
Pharmacokinetics
Tamsulostad is completely and rapidly absorbed from the gastrointestinal tract.
Absorption may be inhibited by the presence of a large amount of food in the stomach. The kinetics of the drug is linear.
The maximum possible content of the active component in the blood serum is detected after six hours.
The connection with plasma proteins reaches 99%. Volume distribution is small – no more than 0.2 liter/kg.
Tamsulostad does not exhibit a first-pass effect. The drug component undergoes slow metabolism in the liver, where active products are formed that retain increased selectivity for alpha 1 -adrenoreceptors. Most of the drug component is in the bloodstream in unchanged form.
Tamsulostad is excreted by the kidneys: 9% of the drug is excreted unchanged. The half-life of a single dose of Tamsulostad is 10 hours.
Dosing and administration
Unless otherwise prescribed by your doctor, the standard dose of Tamsulostad is one capsule daily. The best time to take the medication is in the morning, immediately after eating.
The capsule is swallowed whole with 150-200 ml of water. The capsule should not be broken or chewed.
The duration of the treatment course is determined by the attending physician.

Use Tamsulostada during pregnancy
Tamsulostad is not used to treat females, so taking the medication during pregnancy is also not possible.
Contraindications
Tamsulostad should not be used:
- with an increased likelihood of developing hypersensitivity reactions to Tamsulostad and the active component tamsulosin;
- orthostatic hypotension;
- severe liver damage;
- childhood.
Tamsulostad is not used to treat female patients.

Side effects Tamsulostada
In some patients, treatment with Tamsulostad is accompanied by certain side effects:
- clouding of consciousness, headaches, dizziness;
- increased heart rate;
- lowering blood pressure;
- runny nose, nosebleeds;
- dyspeptic symptoms, thirst;
- allergic reactions;
- episodes of retrograde ejaculation;
- asthenic syndrome.
Overdose
Overdose of Tamsulostad may be accompanied by low blood pressure. If this happens, it is necessary to take measures to facilitate the functioning of the cardiovascular system. The patient should lie down. Additionally, it is necessary to ensure an influx of fresh air.
In severe situations, infusion treatment is carried out using vasoconstrictor drugs.
When taking large amounts of Tamsulostad, it is very important to monitor kidney function and support all vital body functions.
Hemodialysis is not considered effective in this case.
Often, the patient is helped by cleansing the stomach and intestines and taking sorbent agents.
Interactions with other drugs
The combination of Tamsulostad and other alpha 1 -blockers may lead to a decrease in blood pressure.
Tamsulostad should be used with extreme caution in combination with CYP3A4 inhibitors.
Taking Tamsulostad in combination with Paroxetine and Ketoconazole may result in an increase in Cmax values, which, however, is of no clinical significance.
The rate of elimination of the drug may be increased by the action of Diclofenac and Warfarin.
No other significant interactions between Tamsulostad and other medications have been reported.

Storage conditions
Tamsulostad capsules are stored in dark, dry rooms with a temperature range from +15°C to +25°C.
Children's access to areas where medicines are stored should be strictly limited.

Shelf life
Tamsulostad can be stored for up to 3 years in undamaged packaging. The medication is available on prescription.
