Pharmacodynamics
Selegeline is a selective, irreversible MAO inhibitor that destroys dopamine. It acts by reducing the level of dopamine metabolism, thereby preventing it from increasing in neurons.
When combined with Levodopa, the drug potentiates and prolongs its action, while also reducing side effects. As a result, the dosage of Levodopa can be significantly reduced. But if a person's treatment effectiveness does not depend on the dosage of Levodopa, he should not be prescribed Selegiline.
Pharmacokinetics
For maximum bioavailability of the active component, it is necessary to eat food. When Selegiline enters the gastrointestinal tract, it is quickly absorbed. Up to 94% of the substance in a bound form enters the blood serum. Its greatest amount is formed within an hour or two.
Penetrating into the brain and cerebrospinal fluid, the drug and its derivatives do not accumulate in tissues even with prolonged use.
Forming three main derivatives: Selegin - desmethylsegelin, L-amphetamine - L-methamphetamine, the metabolic process occurs in the liver with a characteristic first-pass effect.
Up to 10-85% is excreted through the urinary system, the rest is excreted through the intestines.

Dosing and administration
The tablet should be taken orally immediately after meals in the first half of the day, with a small amount of liquid.
When using Selegin alone or in therapy with Decarboxylase and Levodopa, the initial dose should not exceed 5-10 mg/day. It is prohibited to take more than 10 mg per day.
After starting a combined treatment regimen with Levodopa, it is possible to reduce the amount of the second drug to a minimum.
The daily dose can be divided into two doses.
The attending physician will determine the treatment regimen and duration, taking into account the effectiveness of the drug.
Contraindications
Seliglin should not be combined with Levodopa for patients with thyrotoxicosis, pheochromocytoma, closed-type glaucoma, suffering from high blood pressure and rapid heartbeat. And also when diagnosing such diseases as benign hyperplasia, angina pectoris and progressive dementia.
Refusal of therapy with Selegiline will be the presence of a peptic ulcer in the patient, disorders of the excretory functions of the urinary system and liver.
In addition, the medication is contraindicated in the presence of individual intolerance to any component of the drug.
The drug is not used in children.
Overdose
If the patient complains of dizziness, headache, insomnia or, conversely, drowsiness, weakness throughout the body, anxiety, chest pain - these may be symptoms of excessive consumption of Selegiline in a dose that exceeds 60 mg / day. Also, a health worker may note increased or decreased blood pressure, tachycardia, arrhythmia, confusion, a specific posture with a thrown back head and arched back, spasm of the masticatory muscles.
Symptomatic therapy is used for treatment, since there is no antidote.
Interactions with other drugs
When treating a patient with drugs that act as serotonin agonists, narcotic analgesics, monoamine oxidase inhibitors, as well as drugs such as Paroxetine, Citalopram, Sertraline and Fluvoxamine, the use of Selegiline is strictly prohibited.
You should not drink alcohol while undergoing treatment with Selegiline. After undergoing treatment with this drug, you can start taking Sumatriptan only 24 hours after finishing taking the pills. As for taking drugs that block the reuptake of serotonin, you should wait a period of fourteen days.
Seligilin can be prescribed to a patient after discontinuing serotonin reuptake inhibitors no earlier than seven days later; in the case of taking Fluoxetine, this period will increase to five weeks.
It is not recommended to simultaneously treat Selegiline with drugs that are sympathomimetics.
It should be taken into account that the combined use of this medication and Levodopa potentiates and increases the period of action of the latter. And in combination with the anticholinergic group, there is a possibility of increasing the frequency of side effects.
