Indications Parieta
In tablets, the dosage of which is 10 mg, it is used to treat dyspepsia caused by high gastric pH, which is accompanied by sour belching with heartburn.
The medicine in a dose of 20 mg is used in the following cases:
- aggravated ulcers in the gastrointestinal tract;
- GERD of an ulcerative or erosive nature;
- GERD of non-erosive nature;
- supportive treatment for GERD;
- gastrinoma, as well as conditions observed against the background of pathogenic hypersecretion;
- eradication of Helicobacter pylori bacteria in people with ulcers.

Pharmacodynamics
The element rabeprazole is an antisecretory substance, which is a derivative of benzimidazole. It inhibits the processes of secretion of gastric juice, specifically slowing down the activity of H + /K + -ATPase on the secretory surface of parietal glandulocytes inside the gastrointestinal tract.
H + /K + -ATPase is a protein complex that operates by the proton pump mechanism. This allows us to conclude that rabeprazole sodium is a gastric proton pump inhibitor that blocks the final stage of acid production. This effect is considered to be portion-dependent and causes inhibition of stimulated and basal acid production processes (the irritant bacteria are not important).
Rabeprazole sodium does not have an anticholinergic effect.
Pharmacokinetics
The absorption of the substance from the gastrointestinal tract is carried out at a fairly high speed. Due to this, the Cmax values of the drug are achieved after 3.5 hours (in the case of using a dosage of 20 mg). Absolute bioavailability indicators are about 52%. Multiple use of the drug does not change the bioavailability values.
Protein synthesis of rabeprazole inside plasma is 97%. Most of the substance is excreted in urine (mainly in the form of metabolic products - mercapturic acid conjugate, as well as carboxylic acid and other elements). The remainder of the drug is excreted with bile and feces.
Dosing and administration
The tablets are taken orally, swallowed whole. It should be noted that the activity of rabeprazole does not change in any way from food intake, as well as the time of drug administration.
The medicine in a dosage of 10 mg is taken in the amount of 1 tablet per day (it is recommended to do this in the morning before meals, because this enhances its therapeutic effectiveness).
Tablets of 20 mg are used in case of gastrointestinal diseases occurring with complications, which is why they can be used for various disorders only with the prescription of a specialist. In addition, for different pathologies, not only the dosage regimen and therapy regimen are selected, but also its duration. In this regard, it is prohibited to use Pariet independently, without a doctor's prescription.
Use Parieta during pregnancy
It is prohibited to use Pariet during lactation or pregnancy.
Contraindications
It is contraindicated to use the drug if there is intolerance to rabeprazole.

Side effects Parieta
Available clinical data allow us to conclude that the medication is often tolerated without complications. The side effects that appear are often mild and disappear fairly quickly. These include bloating, dry mouth, headaches or abdominal pain, rash, dizziness, constipation, peripheral edema, etc.
Overdose
Previously, there were no cases of poisoning with Pariet, but the possibility of its development cannot be completely ruled out. It is believed that when using large doses of drugs, the degree of expression of negative symptoms may be potentiated.
When disorders occur, symptomatic procedures are performed.

Interactions with other drugs
Often, the use of this drug in combination with other drugs does not cause significant therapeutic interactions. But it should be taken into account that rabeprazole sodium has a long-term and stable inhibitory effect on the process of gastric juice production. This can lead to the development of interactions with elements whose absorption depends on pH values.
Combination with ketoconazole leads to a decrease in its absorption by 30%, while for digoxin this indicator increases by 22%. In this regard, it is necessary to adjust the doses of these drugs downwards or upwards.
Combined use of the drug with fatty foods slows its absorption by 4+ hours, without affecting its indicators in the body. Combining the drug with methotrexate, especially in large portions, increases the indicators of the latter (or the indicators of its metabolic product, hydroxymethotrexate) almost twofold.
Shelf life
Pariet is allowed to be used within 24 months from the date of manufacture of the therapeutic agent.

Application for children
The medicine in a 10 mg dose is prohibited for use by children under 12 years of age, and in a 20 mg dose – under 18 years of age.

The drug's analogues include such drugs as Famotidine, Rabeprazole, Ranitidine with Omeprazole, as well as Nexium and others.

Pariet is often discussed in light of its therapeutic effectiveness. It is known that the drug is used to treat ulcers, gastritis, heartburn, etc. It is noted that even a temporary decrease in acidity helps restore the gastrointestinal tract.
But not all reviews from those who used this medicine are positive. There are comments that indicate that therapy with its use did not produce results - heartburn and pain did not disappear. Experts believe that the cause may be an incorrectly established diagnosis or an incorrectly selected regimen for using the drug.
