Lacipil is a Ca channel blocker. It belongs to the category of derivatives of the substance dihydropyridine.

Indications Lacipila
It is used for monotherapy in case of increased blood pressure. It can also be used in complex treatment of this disorder – in combination with other antihypertensive drugs: diuretics, β-blockers and ACE inhibitors.
Release form
The drug is released in the form of tablets of 2 and 4 mg. There are 7 tablets inside the blister cell. There are 2 or 4 blister packs in a pack.
Pharmacodynamics
The substance lacidipine has the ability to expand peripheral arterioles, and at the same time lowers blood pressure and reduces total peripheral vascular resistance. The use of medicinal doses of the drug leads to stabilization of the intermembrane current of Ca2+. It should be noted that this does not affect venous tone, and there is no effect on the sinoatrial or atrioventricular nodes. The drug does not have a negative inotropic effect. When using 4 mg of the drug, a slight change in ECG data may be observed - prolongation of the QT interval.
Lacipil exerts an antihypertensive effect by selectively influencing the functioning of Ca channels within vascular smooth muscles.
Pharmacokinetics
Taking the drug orally leads to rapid but weak absorption of lacidipine in the gastrointestinal tract (the absolute bioavailability index is a maximum of 10%). Then the substance undergoes a metabolic process (first liver pass), during which its decay products are formed (there are 4 of them), which have a weak medicinal effect.
Maximum values of LS inside plasma are observed after 30-150 minutes. The component is more than 95% synthesized with α-glycoprotein, as well as albumin.
Excretion occurs in the form of decay products: 70% of the substance is excreted in feces, and the remainder is excreted in urine. After reaching equilibrium values, the half-life of lacidipine is about 13-19 hours.
Dosing and administration
The tablets are taken once a day, at the same time. It is recommended to do this in the morning, regardless of the diet. The medicine is washed down with liquid. It is forbidden to use grapefruit juice for this, because it reduces the level of bioavailability of drugs from the dihydropyridine category.
Sizes of initial and medicinal dosages.
The initial daily dose cannot exceed 2 mg per use, but it can be increased to 4 mg later, and even later to 6 mg. Doses are often adjusted until the desired medicinal effect is achieved (about 3-4 weeks). Doses may vary depending on the severity of the disease and the patient's tolerance of the treatment.

Use Lacipila during pregnancy
There is currently no information regarding the safety of prescribing Lacipil to pregnant women. Animal tests have shown no teratogenic effects or slowing of fetal growth and development. They also showed that the drug and its metabolites can pass into breast milk.
The drug is permitted to be used by pregnant and lactating women only in situations where the benefit to them is greater than the possibility of complications in the fetus or infant.
It is also necessary to take into account the possibility of relaxation of the uterine muscles.
Contraindications
Contraindications include: hypersensitivity to any component of the drug, as well as age under 18 years.
Caution is required in the following disorders:
- disorders in the functioning of the sinoatrial or atrioventricular node;
- prolongation of the QT interval (acquired or congenital);
- poor cardiac output (eg, due to aortic stenosis);
- unstable angina;
- recent myocardial infarction;
- problems with liver function;
- treatment with drugs that prolong the QT interval.
Side effects Lacipila
As a result of peripheral vasodilation, mild adverse effects are observed: hot flashes, heart palpitations, headaches, peripheral edema, and dizziness. These disorders are temporary and often resolve on their own, without stopping treatment using the previously prescribed doses.
Rarely observed side effects include rash, nausea, asthenia, itching with erythema, and in addition, gingival hyperplasia, polyuria and problems with the stomach.
Disorders such as increased plasma levels of the alkaline phosphatase element occasionally occur.
At the initial stage of treatment, the following disorders may be observed: exacerbation of existing angina (especially often observed in individuals with a severe form of coronary heart disease).
Overdose
In case of poisoning, the following symptoms are observed: tachycardia and a marked decrease in blood pressure. Less frequently, bradycardia is observed or there is a slowing of AV conduction.
Symptomatic treatment procedures are used to eliminate the disorders. The medicine does not have a special antidote.
Interactions with other drugs
Lacipil in combination with other antihypertensive drugs (beta-blockers, diuretics and ACE inhibitors) potentiates the antihypertensive properties of the drugs.
Combination with estrogens or NSAIDs, on the contrary, leads to a decrease in the antihypertensive effect of the drug - they block the processes of PG binding inside the kidneys, as well as the retention of Na+ elements.
Plasma lacidipine levels are increased when combined with cimetidine.
Storage conditions
Lacipil is kept in standard conditions for medicines. Temperature – no more than +30°C.
