Indications of the lazipila

It is used for monotherapy with increased blood pressure. It can also be used in the complex treatment of this disorder - in combination with other antihypertensive drugs: diuretics, β-blockers and ACE inhibitors.

Release form

The release occurs in the form of tablets of 2 and 4 mg. Inside the blister cell contains 7 tablets. In a pack - 2 or 4 blister packs.

Pharmacodynamics

The substance lacidipine has the ability to expand the peripheral arterioles, and at the same time lowers blood pressure and reduces OPSS. The use of medicinal dosages of the drug leads to stabilization of the intermembrane current of Ca2 +. It should be noted that in this case, there is no effect on venous tone, and also with no effect on sinus-atrial or atrioventricular nodes. The medicine does not have a negative inotropic effect. With the use of 4 mg of the drug, there may be a slight change in the ECG data - prolongation of the QT-interval.

Latsipil has an antihypertensive effect, selectively affecting the work of the Ca channels inside the smooth muscles of the vessels.

Pharmacokinetics

Taking the drug inside leads to a fast, but weak absorption of lacidipine inside the digestive tract (the absolute bioavailability is a maximum of 10%). Further, the substance passes through the process of metabolism (the first hepatic passage), at which its decay products (their 4) are formed, which have a weak medicinal effect.

Maximum values of LS inside the plasma are observed after 30-150 minutes. A component of more than 95% is synthesized with α-glycoprotein, as well as albumin.

Excretion is carried out in the form of decay products: 70% of the substance - with feces, and the remainder - with urine. After reaching equilibrium values, the half-life of lacidipine is about 13-19 hours.

Dosing and administration

Tablets are drunk once a day, at the same time. It is recommended that you do this in the morning, not taking into account the diet. Drink with a liquid. It is forbidden to use grapefruit juice for this, because it reduces the level of bioavailability of drugs from the dihydropyridine category.

The sizes of initial and medicinal dosages.

The initial daily dose can not exceed 2 mg per 1 use, but it is allowed to further increase to 4, and even later - up to 6 mg. Often, the dosages are selected until the desired medication effect is obtained (about 3-4 weeks is needed). Portions may differ depending on the severity of the disease, as well as the tolerability of treatment by the patient.

Use of the lazipila during pregnancy

Now there is no information regarding the safety of prescribing to pregnant women of Lacipila. Animal tests revealed the absence of a teratogenic effect or a slowing in the growth and development of the fetus. They also showed that the drug with its metabolites is able to pass into the mother's milk.

The drug is allowed to use pregnant and nursing exclusively in situations where the benefits for them are higher than the possibility of complications in the fetus or baby.

It is also necessary to consider the possibility of relaxing the uterine musculature.

Contraindications

Among the contraindications: increased sensitivity to any component of drugs, as well as age under 18 years.

Caution is required for the following violations:

• disorders in the work of the sinus-atrial or atrioventricular node;
• Prolongation of QT-interval (acquired or congenital);
• weak cardiac output (eg, due to stenosis of the aortic orifice);
• an unstable form of angina pectoris;
• recent myocardial infarction;
• problems with hepatic function;
• treatment with the use of drugs prolonging the QT-interval.

Side effects of the lazipila

As a result of peripheral vascular dilatation, slight negative effects are observed: hot flashes, palpitations, headaches, peripheral edema and dizziness. These violations are temporary and often go away by themselves, without discontinuing treatment using previously prescribed portions.

Occasionally, there are such side effects: rashes, nausea, asthenia, itching with erythema, and in addition gingival hyperplasia, polyuria and stomach problems.

Single disorders arise, such as an increase in the plasma indices of the AF element.

At the initial stage of treatment, such violations can occur: exacerbation of existing angina pectoris (in particular, it is often noted in people with severe form of ischemic heart disease).

Overdose

When poisoning, the following symptoms are noted: tachycardia and marked decrease in blood pressure. Less often observed bradycardia or there is a slowing of AV-conduction.

Symptomatic treatment procedures are performed to correct the violations. There is no special antidote for the drug.

Interactions with other drugs

Latsipil in combination with other antihypertensive medications (β-adrenoblockers, diuretics and ACE inhibitors) potentiates the antihypertensive properties of the drugs.

The combination with estrogen or NSAID, on the contrary, leads to a decrease in the antihypertensive effect of drugs - they block the processes of binding of PG within the kidneys, as well as the delay of Na + elements.

The level of lacidipine inside the plasma increases with combination with cimetidine.
