Jintropin is a STH preparation that has a stimulating effect on growth processes.

Indications Jintropina

It is used to treat the following disorders:

  • osteoporosis;
  • slowing down of growth processes in a child observed against the background of a deficiency of STH;
  • a failure affecting renal function in which growth retardation is recorded;
  • Prader-Willi syndrome;
  • severe immunodeficiency accompanied by weight loss;
  • in tissue regeneration and healing of burn areas in the period after surgical procedures;
  • Ullrich syndrome;
  • as HRT for adults.

Pharmacodynamics

Jintropin is a biosynthetic somatotropin created using genetic engineering technologies. The STH gene is introduced into bacterial DNA (E. coli), and then the bacteria promote the synthesis of full-fledged somatotropin.

The composition of the drug and its effect are identical to human STH produced by the pituitary gland. It is a polypeptide containing 191 amino acids. It promotes skeletal growth stimulation – by influencing the epiphysis of tubular bones. In addition, the drug activates the processes of collagen binding to chondroitin sulfate.

It has anabolic properties – it increases the speed of amino acid delivery into cells, as well as protein binding from them. It increases the volume of muscle cells, due to which muscle mass grows.

Along with this, the drug affects fat metabolism: reduces the volume of adipose tissue. It also promotes the retention of mineral salts with nitrogen and liquid in the body. Increases blood sugar levels, inhibiting insulin production.

Pharmacokinetics

The medicinal effect lasts within 12-48 hours.

Dosing and administration

Jintropin should be administered subcutaneously, once a day, at night. The injection sites should be constantly changed to prevent lipoatrophy. The lyophilisate should be diluted exclusively in the solvent supplied with it. The finished medicine has a transparent shade. If the liquid contains particles of undissolved powder or is cloudy, it is prohibited to use it for injection.

It is necessary to select the doses taking into account the degree of STH deficiency, therefore the regimen and duration of drug use should be prescribed exclusively by an endocrinologist.

For a child, daily doses of 0.07-0.1 IU/kg are prescribed. Therapy should be started at an early age and then continued for several years until the onset of puberty (or until the epiphyses of tubular bones close).

In Ullrich syndrome and renal failure complicated by growth retardation, the drug is used in a daily dosage of 0.14 IU/kg. The dose should be increased in the 2nd year of the therapeutic cycle. If the growth dynamics are insufficient, the dosage should be increased again in the 1st year of therapy.

An adult is initially prescribed a dose of 0.45-0.9 IU per day, then increasing it based on the effect produced.

Use Jintropina during pregnancy

It is prohibited to use Jintropin during pregnancy.

Contraindications

Main contraindications:

  • the presence of severe sensitivity to the drug;
  • neoplasms in the brain;
  • closure of bone epiphyses.

At the same time, caution is needed when prescribing to people with hypothyroidism, as well as with diabetes mellitus or increased intracranial pressure.

Side effects Jintropina

The administration of the drug may cause the following side effects:

  • headaches;
  • hyperglycemia;
  • feeling tired or weak;
  • increased intracranial pressure (symptoms include vomiting, visual disturbances, nausea and, along with this, headaches);
  • the appearance of hypothyroidism;
  • leukemia;
  • the appearance of hyperemia, and in addition itching or severe swelling in the injection area, as well as the development of lipoatrophy (a decrease in the volume of adipose tissue);
  • destruction of the integrity of the cartilage in the area of the femoral head;
  • the appearance of rashes on the epidermis;
  • fluid retention.

Such manifestations are temporary and depend on the size of the drug dose.

Less common negative symptoms include: pancreatitis, gynecomastia, swelling in the optic disc area, hearing impairment, accelerated growth processes, hip subluxation in a child, and in addition, progression of scoliosis and malignancy of a nevus that the patient already had.

Overdose

Poisoning causes first hypo- and then hyperglycemia. Constant prolonged intoxication provokes the appearance of hypothyroidism, acromegaly, and gigantism.

Interactions with other drugs

GCS substances lead to a weakening of the therapeutic properties of STH.

Estrogens, anabolics and thyroid hormones also affect the medicinal effectiveness of the drug.

Jintropin must be kept at a temperature of 2-8°C. The finished medicinal substance can be kept at a temperature of 2-8°C for a period of 14 days.

Shelf life

Jintropin can be used within 36 months from the date of release of the therapeutic agent.
