Pharmacodynamics
Due to its slightly alkaline properties, the active substance of the drug Gasec, omeprazole, covalently binds to H+/K+-ATPase, a hydrolase enzyme, potassium-hydrogen adenosine triphosphatase (proton pump), which catalyzes the synthesis of hydrochloric acid. The activity of this enzyme is inhibited, as a result of which the parietal cells of the gastric mucosa reduce the production of HCl, reducing the acidity of gastric juice (increasing its pH). This helps to stop pain and heartburn in hyperacid diseases of the gastrointestinal tract.
Pharmacokinetics
After oral administration of Gasec, the active substance (omeprazole) is absorbed in the stomach and small intestine; its bioavailability is 40%. Plasma protein binding is almost 97%; the highest plasma concentration is achieved 4 hours after administration of the drug.
Biotransformation of the active substance occurs in the liver; metabolites are excreted in urine and feces - with a half-life of omeprazole of about one hour.
Dosing and administration
Gasek capsules are taken orally (both before and after meals), washed down with water. The daily dose is one capsule. The standard course of administration is one to two months. However, if the drug does not show any effectiveness within the first week, the use of Gasek should be discontinued.
Use Gasek during pregnancy
Although clinical studies of the use of omeprazole in pregnant women have not revealed any negative effects on the fetus, Gasec capsules are not prescribed to reduce heartburn during pregnancy.
Side effects Gasek
Possible side effects of Gasec include:
- nausea, vomiting, dry mouth, intestinal dysfunction, increased gas formation, abdominal pain, jaundice;
- increased temperature, bronchospasm, allergic bullous rashes on the mucous membranes and skin (Stevens-Johnson syndrome);
- decrease in the level of leukocytes, platelets and agranulocytes in the blood;
- decrease in the content of Na and Mg in blood plasma;
- muscle and joint pain;
- headaches, dizziness, blurred vision, insomnia, paresthesia, a state of unreasonable anxiety and depression.
Interactions with other drugs
Gasec is not used simultaneously with antiviral drugs for the treatment of HIV (Reyataz, Viracept, etc.). Gasec should not be taken simultaneously with some antitumor drugs (tarlenib, etc.), indirect anticoagulants (phenindione, warfarin, syncoumar), as well as antifungal drugs of the imidazole and triazole groups.
Due to the risk of exceeding the concentration of digitalis alkaloids in the blood plasma, the simultaneous use of Gasek and antiarrhythmic drugs of the cardiac glycoside group (digoxin, lanoxin, novodigal) is not allowed.
Storage conditions
Store at room temperature.
Shelf life
3 years.
