Pharmacodynamics
Anthranoid compounds, which are constituent components of aloe, and in addition inside the crust of the buckthorn, have a laxative effect. This mechanism is due to the effect of drugs on the motility of the colon - it carries out the processes of suppressing spasms, as well as stimulation of propulsive contractions. As a result, there is an acceleration of the passage of feces within the intestine, which also reduces the resorption of the fluid.
Along with this, due to active excretion of chlorides, electrolytes with water are also removed. The laxative effect is observed 6-10 hours after the use of the tablet.
Pharmacokinetics
Anthranoid glycosides that possess a β-glycosidic bond are the original ligaments that can not be absorbed and absorbed in the upper GI tract. Only within the large intestine these elements are transformed with the help of enzymes of the bacterial flora, and also mucous (partially) into free-type aglycons (emodin). These substances are decay products that have a laxative effect.
Only 20-25% of the dose taken inside is absorbed inside the body. The indicator of bioavailability of emodin is significantly lower than that of absorption, because it is rapidly oxidized with conversion to rhein, as well as an unknown degradation product. Inside the serum, the substance reaches a peak level 1.5-3 hours after ingestion. Peak serum values are about three times higher than those within the ovaries, and also ten times in the testicles.
Most of the decay products are transformed inside the cells by intestinal mucosa, and then excreted with feces. The remainder of the substance undergoes the process of resorption and is excreted in the form of sulfates or glucuronides along with urine.
